Today, i went back to the school to coach the junior that will be taking over our projects on the AKTA system....
Besides by Akon's 2 project groups, we also taught one of Mr Wong's group. 3 groups together..... quite a big group.... -.-'
We reached W316 an hour before the juniors arrive.... we prepared the buffer and waited for them.... lol... they came around 20 mins late lah.... no sense of time.... haha.... a bit too strict maybe... we showed them how to use the vacuum filter to filter the buffer, how to use the centrifuge in the lab and how to prepare the crude protein that we will be using in the protein purification process...
It was at around 1.30pm maybe, we finally started on the AKTA system. I started to explain how to use the AKTA system with the aid of Noor... I should have brought the wireless mouse sia... it was so hard to look at the screen from the place that i was standing.... haiz.... The juniors gave a few blank looks when i was explaining the procedures... haha... look like my group 1 year ago when we were listening to Mr Wong's explaination...
They took down a lot of notes.... they copied very fast.... but sometimes i still had to repeat some steps when they din catch wat i said.... after a while, they are starting to lose their concentration. ~.~ can't blame them though, it is a bit too much to digest for one day.... but i guess they will get a hang of it soon... they asked a few questions through out the process and George would answer them... After storing the system in 20% ethanol, George talked about the chromatograph, like how to locate the protein, how to estimated the % of elution buffer etc...
I also taught them how to use the machine that takes the photo of the sds gel... Noor touched on how to prepare the samples for loading into the sds gel and also the staining of the gels...
We ended about 4.30pm i think... when we left, the juniors were still in the lab...
